Description
Cluster of Differentiation 70 (CD70) is also known as CD27 ligand (CD27L / CD27LG), TNFSF7, TNFSF7G, is a type II transmembrane glycoprotein belonging to the TNF superfamily (TNFSF) and is a surface antigen found on activated T-and B-lymphocytes and mature dendritic cells. Binding of CD70 to its receptor CD27 induces in priming, effector functions, differentiation and memory formation of T-cells, and thus is invloved in the biological processes including T-cell activation, the proliferation of costimulated T-cells, as well as the generation of cytolytic T-cells. CD70 on T cells provides costimulatory signals that are required for T cell proliferation, clonal expansion and the promotion of effector T cell formation. CD70 on mouse B cell has been shown to inhibit terminal differentiation of activated B cells into plasma cells and enhances commitment to memory B cell responses. CD70 induces proliferation and IFNγ production, on NK cells.

Assay Principles
This inhibitor screening ELISA kit is designed to facilitate the identification and characterization of new CD27L pathway inhibitors. The assay takes advantage of our in house-developed binding of biotinylated human CD27L to immobilized human CD27 in a functional ELISA assay, and employs a simple colorimetric ELISA platform. Briefly, we provide you with a human Biotinlated CD27L protein, a human CD27 protein, an anti-CD27L neutralizing antibody (as method verified Std.), and Streptavidin-HRP reagent.
Your experiment will include 4 simple steps:
a) Coat the plate with human CD27.
b) Add your molecule of interest to the tests.
c) Add human CD27L-Biotin to bind the coated human CD27.
d) Add Streptavidin-HRP followed by TMB or other colorimetric HRP substrate.
Finally, the half maximal inhibitory concentration (IC50) of your compound to CD27L: CD27 binding will be determined by comparing OD readings among different experimental groups.

Serial dilutions of Anti-CD27L Neutralizing antibody (1:1 serial dilution, from 5 μg/mL to 0.005μg/mL) was added into CD27: Biotinlated CD27L binding reactions. The assay was performed according to the protocol described below. Background was subtracted from data points prior to log transformation and curve fitting.
