Recombinant cynomolgus CD8A (XP_001092778.2) (Met42-Asp223) was fused with a flag tag at the C-terminus, constructed the plasmid 1 ; Recombinant cynomolgus CD8B (XP_005575417.1) (Met1-Pro170) was fused with a polyhistidine tag at the C-terminus, constructed the plasmid 2. The two plasmids were co-expressed and the heterodimer was purified.
Description
CD8 exists as a disulfide-linked dimer of either α and β chain or two α chains. The crystal structure of the N-terminal 113 amino acids of human CD8 revealed homology to immunoglobulin (Ig) variable domains with nine β strands divided into two β sheets, one of four and the other of five strands.
