Recombinant human CD8A (NP_001759.3) (Met1-Asp182) was fused with a flag tag at the C-terminus, constructed the plasmid 1; Recombinant human CD8B (NP_004922.1) (Met1-Pro170) was fused with a polyhistidine tag at the C-terminus, constructed the plasmid 2. The two plasmids were co-expressed and the human CD8A&CD8B heterodimer was purified.
Description
CD8 exists as a disulfide-linked dimer of either α and β chain or two α chains. The crystal structure of the N-terminal 113 amino acids of human CD8 revealed homology to immunoglobulin (Ig) variable domains with nine β strands divided into two β sheets, one of four and the other of five strands.
