Storage

Reconstitution

It is recommended that sterile water be added to the vial to prepare a stock solution of 0.2 mg/ml. Centrifuge the vial at 4ºC before opening to recover the entire contents.

Human PD-L2, Mouse IgG1 Fc Tag on SDS-PAGE under reducing (R) condition. The gel was stained overnight with Coomassie Blue. The purity of the protein is greater than 95%.

Bioactivity-ELISA 1

Immobilized Human PD-1, His Tag at 2 ug/mL (100 uL/well) can bind Human PD-L2, Mouse IgG1 Fc Tag with a linear range of 10-156 ng/mL.

Bioactivity-ELISA 2

Serial dilutions of nivolumab were added into Human PD-L2, Mouse IgG1 Fc Tag: Biotinylated Human PD-1, Fc,Avitag binding reactions. The half maximal inhibitory concentration (IC50) is 0.2823 ug/mL.

Bioactivity-FACS 1

Flow Cytometry assay shows that Human PD-L2, Mouse IgG1 Fc Tag can bind to 293 cell overexpressing human PD-1. The concentration of PD-L2 used is 0.1 ug/mL.

Bioactivity-FACS 2

FACS analysis shows that the binding of Human PD-L2, Mouse IgG1 Fc Tag to 293 overexpressing PD-1 was inhibited by increasing concentration of neutralizing Anti-PD-1 antibody. The concentration of PD-L2 used is 0.1 ug/mL. The IC50 is 1.3 ug/mL.
