Storage

Reconstitution

It is recommended that sterile water be added to the vial to prepare a stock solution of 0.2 mg/ml. Centrifuge the vial at 4ºC before opening to recover the entire contents.

Human SIRP alpha, Mouse IgG1 Fc Tag (HPLC-verified) on SDS-PAGE under reducing (R) condition. The gel was stained overnight with Coomassie Blue. The purity of the protein is greater than 95%.

SEC-MALS 1

The purity of Human SIRP alpha, Mouse IgG1 Fc Tag was more than 90% and the molecular weight of this protein is around 150-165 kDa verified by SEC-MALS.

Bioactivity-ELISA 1

Immobilized Human CD47, His Tag at 2 ug/mL (100 uL/well) can bind Human SIRP alpha, Mouse IgG1 Fc Tag (HPLC-verified) with a linear range of 4-31 ng/mL.

Bioactivity-ELISA 2

Serial dilutions of Anti-Human CD47 Neutralizing Antibody were added into Human SIRP alpha, Mouse IgG1 Fc Tag: Biotinylated Human CD47, Fc,Avitag binding reactions. The half maximal inhibitory concentration (IC50) is 0.2006 ug/mL.

Bioactivity-FACS 1

FACS assay shows that Human SIRP alpha, Mouse IgG1 Fc Tag can bind to Jurkat cell expressing CD47. The concentration of SIRP alpha used is 0.3 ug/mL.

Bioactivity-FACS 2

FACS analysis shows that the binding of Human SIRP alpha, Mouse IgG1 Fc Tag to Jurkat expressing CD47 was inhibited by increasing concentration of neutralizing anti-CD47 antibody. The concentration of SIRP alpha used is 0.3 ug/mL. IC50=0.1318 ug/mL.
