Description

SP140 nuclear body protein (SP140) is a component of the nuclear body that may be involved in trafficking between the nucleus and the cytoplasm. SP140 contains one HSR domain, one PHD-type zinc finger, a bromodomain and one SAND domain. The bromodomain functions as a 'reader' of epigenetic histone marks and regulates chromatin structure and gene expression by linking associated proteins to the recognized acetylated nucleosomal targets. SP140 may be involved in the pathogenesis of acute promyelocytic leukemia and viral infection.

HTRF Assay for Recombinant SP140 (688-862) protein activity. 3 μM histone peptide H3K14ac was incubated with SP140 (688-862) in reaction buffer including 50 mM HEPES-NaOH pH 7.0, 0.1% BSA for 1 hour at room temperature. Anti-FLAG antibody was used to detect reaction products.
