Recombinant Human SP140 Protein (688-862 aa), His/FLAG-tagged
Product Description
Cat
IMP-6466
Official Symbol
SP140
Product Overview
The peptide corresponding to amino acids 688-862 that contains the bromodomain sequences of SP140 (accession number NP_009168.4) was expressed in E. coli and contains an N-terminal His-Tag and C-terminal FLAG-Tag with an observed molecular weight of 26.98 kDa. The recombinant protein is ≥ 90% pure by SDS-PAGE.
Description
SP140 nuclear body protein (SP140) is a component of the nuclear body that may be involved in trafficking between the nucleus and the cytoplasm. SP140 contains one HSR domain, one PHD-type zinc finger, a bromodomain and one SAND domain. The bromodomain functions as a 'reader' of epigenetic histone marks and regulates chromatin structure and gene expression by linking associated proteins to the recognized acetylated nucleosomal targets. SP140 may be involved in the pathogenesis of acute promyelocytic leukemia and viral infection.

Recombinant proteins in solution are temperature sensitive and must be stored at -80 centigrade to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
SDS-PAGE
SDS-PAGE

SP140 (688-862) protein was run on a 12.5% SDS-PAGE gel and stained with Coomassie Blue. MW: 26.9 kDa Purity: > 90%

Bioactivity-ELISA 1
Activity 1

HTRF Assay for Recombinant SP140 (688-862) protein activity. 3 μM histone peptide H3K14ac was incubated with SP140 (688-862) in reaction buffer including 50 mM HEPES-NaOH pH 7.0, 0.1% BSA for 1 hour at room temperature. Anti-FLAG antibody was used to detect reaction products.

Bioactivity-ELISA 2
Activity 2

HTRF Assay for Recombinant SP140 (688-862) protein activity. 3 μM histone peptide H3K14ac was incubated with SP140 (688-862) in reaction buffer including 50 mM HEPES-NaOH pH 7.0, 0.1% BSA for 1 hour at room temperature. Anti-FLAG antibody was used to detect reaction products.
